​Facilities​

Our group apply a variety of techniques to study the biogeochemistry of the ocean and the sensitivity/response of marine organisms to the environment.   We use a variety of techniques based on fluorescence, spectrophotometry, chromatography, HPLC, mass spectroscopy, and sequencing, the lasts ones with the support of the Analytical Core Lab and the Bioscience Core Lab​, both at KAUST.

​​PHYTO-PAM

PHYTO-PAM instruments facilitate measurements of mixed algae populations for distinct analysis of their photosynthetic performance and light-adaptation states. Green algae, diatoms, cyanobacteria.
This equipment​ include:
  • Power-and-Control-Unit to connect the LED-Array-Cones PHYTO-ML and PHYTO-AL, the Photomultiplier-Detector PM-101P, the Miniature Magnetic Stirrer PHYTO-MS and the Spherical Micro Quantum Sensor US-SQS.
  • Emitter-Detector Unit to be connected to Power-and-Control-Unit PHYTO-C; featuring special Circular LED-Array for fluorescence excitation with blue (470 nm), green (520 nm), orange-red (645 nm) and red light (665 nm) as well as for actinic illumination with continuous red light up to 2000 μmol quanta m-2s-1 PAR.
  • Stirring Device to be mounted on Emitter-Detector Unit Spherical Micro Quantum Sensor consisting of 3.7 mm diffusing sphere coupled to integrated PAR-sensor via 2 mm fiber

Microscopy-PAM

The MICROSCOPY-PAM is an extremely sensitive non-imaging chlorophyll fluorometer specialized to measure smallest spots in microscopic specimen like tissue preparations or suspensions.

Essentially, the fluorometer consists of a modified epi-fluorescence microscope equipped with a modulated LED light source and a photomultiplier for detection of modulated chlorophyll fluorescence.

The MICROSCOPY-PAM system uses a Zeiss AxioScope.A1 epifluorescence microscope. In our equipment, the microscope is equipped with 3 wide-aperture Zeiss Fluar objective 20X, 40X and 100X magnification. Standard light source for fluorescence excitation is the blue Zeiss LED-Module 470 nm and also it has the UV LED-Module 365 nm.

Fluorescence excitation and detection are controlled by the PAM-CONTROL unit which allows stand-alone operation of the MICROSCOPY-PAM but can also function as a physical interface for computer-controlled operation of the system.

TRILOGY Laboratory Fluorometer

The Trilogy make fluorescence, absorbance and turbidity measurements using the appropriate snap-in Application Module. It has excellent sensitivity and a wide measurement range. The all solid-state design ensures reliable and repeatable readings for many years.
Fluorescence modules that our group has available are:
  • Extracted chlorophyll a non-acidification.
  • Tracer Dyes rhodamine, fluorescein and PTSA.
  • CDOM/FDOM, refined and crude oil, optical brighteners, histamine and ammonium.

LS55 Fluorometer

The LS55 is a monochromator-based fluorescence spectrometer which uses a high energy pulsed xenon source for excitation. Xenon minimizes photobleaching of samples and provides a long-lived excitation source. The variable slit and holographic gratings provide flexibility with very low stray radiation.

Holographic gratings are also used on both the excitation and emission monochromators to further reduce stray light and improve the system’s performance. The system is also equipped with a reference signal system which provides control over signal dynamics and reduces noise effectively.

The system has FL WinLab™ software which seamlessly combines PerkinElmer’s extensive application-specific knowledge and instrument control with the ease-of-use provided by the Windows® operating environment. Specific modes of instrument operation such as Scan, Time Drive, and Ratio Data Collection can be easily accessed from the application menu of the software.

Excitation and emission monochromators can be independently or synchronously scanned, while the Prescan mode is ideal for method development and locating excitation and emission maxima.

FL WinLab includes a validation protocol that automatically checks the instrument performance to ensure that it is operating within specification. The plate- reader accessory lets you switch from a fully functional fluorescence spectrometer to a well- plate reader in seconds. The plate reader is mounted on the fron t, providing easy access to standard multi -well plates.

LAMBDA 1050 UV/Vis/NIR

LAMBDA 1050 UV/Vis/NIR Spectrophotometer has been designed to achieve a greater level of sensitivity, resolution and speed in the NIR range, it simplifies the analysis of difficult samples such as high absorbing glass, optical coatings, or thin film filters. It features two large sampling compartments and a variety of snap-in modules and accessories, including a general purpose optical bench, integrating spheres, and our patented Universal Reflectance Accessory.
  • Deuterium and Tungsten Halogen light sources with double holographic grating monochromators.
  • PMT, InGaAs, and PbS 3-detector module for testing across the entire UV/Vis/NIR range from 175 nm to 3300 nm.
  • 150 mm Integrating Sphere with PMT and InGaAs detectors (250 nm ∼ 2500 nm) for diffusive reflectance and transmittance measurement. Samples can be mounted inside the sphere.
  • Common beam depolarizer and absolute reflectance accessory. LAMBDA 1050 with its triple detector capability for ultra -high UV/ Vis/NIR performance for wavelengths up to 3300 nm with extreme sensitivity in the NIR region 800-2500 nm and higher energy throughput. For applications such as highly reflective and anti-reflective coatings, all types of glass from clear to highly absorbing safety glass, optical filters of all types from the deep UV through the NIR and many more applications requiring the best photo-dynamic range and the widest possible sampling capability.

FC204 - Fraction Collector
The FC 204 is the ideal large-bed fraction collector, capable of handling a wide variety of collection vessels, including microplates, vials and tubes. The built-in keypad makes set up and operation simple and efficient. Collect fractions by time, drop or peak.
  • In peak mode, collector monitors a detector signal and identifies peaks using either an adaptive-slope algorithm or a specified millivolt threshold.
  • ​​Up to 10 programmable time windows can be added for collecting only what you want, while discarding the column’s void volume, peaks of no interest and equilibration volumes.
  • Stand-alone instrument with an easy-to-use keypad, or can be controlled through your PC via TRILUTION® Software.
  • 3-way diverter valve prevents contamination of collected fractions.
  • Contact closure inputs are available to remotely start/advance or stop—compatible with most HPLC systems.
  • Compact size fits in most fume hoods.
  • Multicolumn adapters for collecting up to 18 separate channels.











MICROX 4 - Oxygenmeter 

​The Microx 4 is a completely stand-alone, portable fiber optic oxygen meter. It can be used with non-invasive sensors & probes (1 mm fiber), and oxygen microsensors (200 µm fiber) in different designs. This oxygen meter is compatible with the wide range of sensors type PSt7 (detection limit 15 ppb, 0 – 100 % oxygen). With its integrated barcode reader the Microx 4 can easily recognize and assign calibration data to sensors in just one scan. The implemented sensor management system allows storing data of up to 100 sensors. 

The main features of this equipment:

​WICROX 4 - Oxygenmeter​​​

WITROX 4 is a four-channel oxygen instrument for use with fiber optic mini sensors (optodes). It has a build-in BlueTooth transmitter for wireless PC communication, and includes a high accuracy temperature sensor and software for compensation of oxygen data to changes in temperature, salinity or barometric pressure in real-time.
The wireless Bluetooth 2.0 communication allows you to keep that vulnerable PC/Notebook at a distance avoiding hazards like water or blood. Also problems associated with USB and serial communication are avoided, and the rugged compact design offers excellent protection and take up minimum bench space. 
The instrument comes with a 4-wire Pt1000 temperature sensor (Class B 1/3 DIN) with  +/- 0.15°C accuracy for 5-digits real-time compensation of oxygen data.
